Scientists at Queen Mary, University of London have uncovered a link between two genes which shows how stem cells could develop into cancer.

The research, published in the online journal PLoS ONE, found a novel mechanism which could be the catalyst for stem cells changing into a tumour.

Dr Ahmad Waseem, a reader in oral dentistry at Queen Mary, University of London who led the research, said: "It was quite an unexpected discovery. We set out to investigate the role of the stem cell gene Keratin K15 which was thought to be a biomarker for normal stem cells.

"Through our research, we discovered there was link between K15 and the notorious cancer gene FOXM1. We found FOXM1 could target K15 to induce cancer formation."

Cancer develops when there is a problem with stem cells; the cells that carry out internal repairs throughout the human body. The loss of stem cell function leads to uncontrolled growth which ultimately develops into a tumour.

The team went through a process where they used extremely sensitive cell and molecular approaches to establish this link.

The study, funded by the Facial Surgery Research Foundation, Saving Faces, paves the way towards identifying new anti-cancer drugs which could be tailored towards cancer stem cells.

Consultant oral and maxillofacial surgeon Professor Iain Hutchison, founder of Saving Faces and co-author on the study, said: "We are excited about this finding as it could lead to more effective cancer drugs being developed to target cancer stem cells and prevent cancer recurrence."

Nora Ephron's final act played out in Manhattan on June 26 where the 71-year-old writer and movie director died from pneumonia brought on by acute myeloid leukemia (AML), one of the most common types of leukemia among adults. AML is a cancer caused when abnormal cells grow inside bone marrow and interfere with the production of healthy blood cells. The marrow eventually stops working correctly, leaving a person with an increased risk of bleeding and infections.

Ephron?best known for writing When Harry Met Sally and Sleepless in Seattle?was first diagnosed in 2006 with one of the myelodysplastic syndromes (MDS), a category of blood diseases also referred to as "preleukemia" that can progress into AML if the bone marrow continually fails to produce enough healthy platelets, red blood cells and white blood cells over time. MDS made headlines recently when ABC's Good Morning America anchor Robin Roberts announced she has been diagnosed with the disease.

Some types of leukemia, including AML, develop as a result of exposure to certain chemicals (including herbicides and pesticides), chemotherapy drugs (such as etoposide and a class of drugs known as alkylating agents) and radiation. Typically, however, a doctor is unable to pinpoint the exact cause in individual cases.

Although estimates vary, there are between 10,000 and 12,000 new cases of MDS in the U.S. annually. More than 80 percent of all MDS patients are older than 60. The National Cancer Institute projects that 13,780 men and women?7,350 men and 6,430 women?will be diagnosed with AML and that 10,200 men and women will die of the malady this year.

Scientific American spoke with Bart Scott, a medical oncologist specializing in the treatment of patients with MDS, about syndrome's progression to AML, who is most at risk for this cancer and whether there are any promising treatments on the horizon. Scott is also director of hematology and hematologic malignancies at the Seattle Cancer Care Alliance and an assistant member of the Fred Hutchinson Cancer Research Center's clinical research division.

[An edited transcript of the interview follows.]

What is the typical trajectory for this form of cancer?
The trajectory of myelodysplastic syndrome is highly dependent upon two factors?the bone marrow myeloblast count and the cytogenetics. [A myeloblast is an immature blood cell that will eventually develop into a type of white blood cell.] The higher the bone marrow myeloblast count is at time of diagnosis the more likely a patient is to progress to acute myeloid leukemia. In general, the more abnormal the cytogenetics [a cell's hereditary and functionalm characteristics] are the more likely a patient is to progress to AML, but there are certain abnormalities that have a good prognosis.

Who are the typical victims of this form of leukemia?
The median age of diagnosis of MDS is 72, it is slightly more common in men than women, and there is a correlation between incidence and age. I would say that Ephron's course does seem consistent with MDS progression to AML.

What are the risk factors for this cancer?
They are based on increasing age as well as certain environmental exposures like pesticides and herbicides. This is not casual contact?we are talking about prolonged, persistent exposure. The amount of exposure required to cause the disease is unknown, however.

Given that Ephron lived most of her life in New York City, whose residents are not typically exposed to inordinately large amounts of pesticides or herbicides, what might account for her condition?
Some researchers have tried to understand MDS in terms of clustering, although there is very little clustering data available. The idea is to understand whether MDS tends to cluster in areas of higher exposure to certain toxic agents. In 2007 Xiaomei Ma [an associate professor of epidemiology the Yale School of Public Health] and a team of researchers published a paper in Leukemia Research regarding this phenomenon. They found that cases of MDS in Connecticut were clustered near the western border. In general, clustering supports the idea that environmental exposure is contributory to the development of a disease. This is the only paper that I know of that has demonstrated clustering in MDS.

We are doing a similar study in the state of Washington. Hanford Site is a nuclear production complex near the Tri-Cities area in southeastern Washington [encompassing Kennewick, Pasco and Richland]. We are looking to see if there is clustering in the Tri-Cities area. But these studies are difficult given the mobility of the area's population, unknown exposure time and any delay in the development of MDS after exposure.

What is the usual treatment?
There are three FDA approved treatments. One is azacitidine, approved by the FDA in 2004 and marketed as Vidaza. Decitabine, sold as Dacogen and approved in 2010 to treat MDS, is another option. [The FDA's oncologic drugs advisory panel in February, however, recommended against the approval of Dacogen to treat older patients with AML. The drug's maker wants FDA approval to use Dacogen in AML patients 65 years and older and who are not considered good candidates for high-dose chemotherapy as an initial treatment for AML.] The third is lenalidomide, also known as Revlimid and introduced in 2004.

However, the only curative treatment is stem cell transplantation. Stem cells are infused just like a blood transfusion. Sources of stem cells include cord blood cells, peripheral blood mobilized stem cells or bone marrow. Success depends on the stage of disease. The one-year treatment related mortality is approximately 20 percent. If patients are transplanted in an early stage of the disease, we have a success rate of 80 percent.

Are there any promising treatments in late-stage testing?
Yes, there are several drugs being investigated in phase II and phase III trials. We currently have a phase III trial open with a drug called rigosertib for patients who have failed prior treatment with azacitidine or decitabine, which are known as demethylating agents. The proposed mechanism of action for these drugs is that they alter gene expression profiles in the cancer cells and increase their susceptibility to death. Rigosertib is a cell cycle inhibitor and would prevent the cancer cells from growing and induce direct damage to the cancer cells, causing their death.

UNC's Barnes has to wait, but all's golden at NBA draft

The Ames, Iowa, native, ranked in the top-five of most mock drafts heading into Thursday night's real deal on the NBA big stage, had to watch six players get their names plastered to the Prudential Center big board before his turn came.

He said he didn't mind the wait. Barnes was very composed and kept it together. The audience reaction was low-key.

"I'm just truly blessed," the 6-8 small forward out of the University of North Carolina said after getting selected by the Golden State Warriors with the seventh pick of the first round. "I don't really have a lot of words right now other than that."

When the Ames High School graduate finally heard his name called by NBA Commissioner David Stern, he bear-hugged his mother, Shirley, and sister Jourdan-Ashle, 13. The women couldn't stop smiling.

"He was calm, he was calm. I think I was pretty composed, as well," his mother said. "But really, just overjoyed. He's just happy to be a Warrior and just happy to actually be a league player now."

After fastening his Warriors' hat, there was only one thing left to worry about for Barnes.

"The biggest thing I was thinking was, 'Don't trip,'" said Barnes, decked out in a conservative yet slick grey suit. "I was just a little nervous and excited that I got picked."

Barnes needed a couple deep breathes after shaking Stern's hand but walked off the stage with a wide, bright smile and a shiny gold and blue Warriors hat to go with it, and was directed straight over to be interviewed on ESPN.

To everyone who congratulated him he said, "Thank you."

He was led through the tunnels to the NBA Entertainment suite for more interviews for at least 30 minutes, then back out to do an interview with Sirius Radio. The big smile never left his face as he took it all in.

North Carolina coach Roy Williams sat with Barnes' mom and sister and said his former Tar Heel star "is going to be a big-time basketball player in the NBA for many years."

"It's a fantastic night for the whole family," Williams said. "He had a great following in Ames, Iowa (where he led Ames High School to a 53-0 record and two state title between 2009-10). He had a great following at Chapel Hill. Harrison's an incredible young man."

Barnes averaged 15.7 and 17.1 points in his two seasons at UNC, becoming one of six Tar Heels to average at least 15 points as a freshman and sophomore.

"It was his goal to be in the NBA, so to have that come to fruition now is special," his mom said.

Barnes was one of 14 players selected to participate in the draft ceremonies ? and enjoy a lunch with Stern at The Westin Hotel in Times Square earlier Thursday, a sure sign of the potential most NBA experts saw in Barnes.

"He can play two positions for (Golden State), he can rebound the basketball. But the best thing he does is, he scores," Williams said. "I've never seen an NBA team that didn't need people who could score. Harrison is a great pick for them."

Barnes had a group of 26 supporting him draft night, including aunts, uncles and cousins who watched him and UNC teammates Kendall Marshall, John Henson and Tyler Zeller follow as first-round selections.

Marshall was taken at No. 13 by the Phoenix Suns, Henson went next to the Milwaukee Bucks and Zeller was taken by the Dallas Mavericks with the 17th pick, with his rights then traded to the Cleveland Cavaliers.

Barnes has never been to the Bay Area but said he looks forward to heading there Friday and starting his career with the Warriors.

"It's a very first-class organization," Barnes said. "I can't wait to get out there and play for them.

"Whatever my ceiling is, I'm definitely going to try to reach it."

Microsoft attack puts spotlight on Greek tensions

ATHENS (Reuters) - Gunmen driving a van packed with gas canisters firebombed the Athens headquarters of Microsoft on Wednesday, underscoring the threat of instability as Greece asks for more time and less hardship in cutting its crippling debt.

Fire gutted the ground floor of the blue four-storey HQ of the U.S. software giant, blackening its walls with flames, on the eve of yet another European summit seeking a solution to a regional debt crisis first unleashed by Greece.

At least two attackers were involved in the sophisticated assault at 4.45 a.m. (0145 GMT) in Maroussi, a northern suburb of the sprawling Greek capital, police said.

Brandishing pistols and an automatic rifle, they kept security guards at bay and set fire to the van carrying three gas canisters and five cans of gasoline. No one was hurt in the early morning assault.

There was no immediate claim of responsibility and police said it was too early to say who was to blame. Suspicion, however, fell on left-wing militants, who have a long and violent history in Greece. Anti-terrorist units were investigating the incident.

Since the end of a military dictatorship in 1974, Greece has been scarred by political violence and assassination, much of it carried out by the militant November 17 group, named after a student uprising which was bloodily suppressed by the army.

Authorities have dismantled the group, whose victims included the CIA station chief in Athens, but splinter organizations committed to the same radical left-wing or anarchist ideals remain active.

Social tensions are rising in Greece over the harsh terms of an international bailout saving the country from bankruptcy but only at the cost of deep economic suffering.

Branches of Macdonald's, Citi and Starbucks in Athens have been set on fire by protesters and militants in the past using petrol bombs or gas canisters.

The attacks usually take place at night and there have rarely been injuries, but three bank staff suffocated in May 2010 when protesters set their branch on fire during a protest march in central Athens.

The assault on Microsoft is a new headache for big multi-nationals in Athens already considering quitting the debt-ridden country because of unpaid bills, falling revenues and the prospect that Greece might be forced to leave the euro.

"They entered the office in a van ... in an effort to burn the whole place down," Ernst-Jan Stigter, general manager of Microsoft's Greek unit, told reporters on Wednesday. "We're grateful there were no injuries and all our crew is safe."

The fire department estimated the damage at about 60,000 euros ($75,000). Staff were told not to come to work.

Contacted by Reuters, Citibank, Coca-Cola and pharmaceuticals giant GlaxoSmithKline said they had no immediate plans to step up security, which has already been tightened after violent protests seen previously in Athens.

NO QUICK RESPITE

The Greek government is unlikely to win any immediate respite from the bailout conditions at a two-day summit of EU leaders starting on Thursday whose chances of making a decisive response to the debt crisis are rated as low.

The harsh conditions attached to a 130-billion-euro ($162.12 billion) lifeline from the European Union and International Monetary Fund, on top of 110 billion in 2010, have helped condemn Greece to five years of record-breaking recession that have enraged the population.

Greeks argue that the cuts in jobs, wages and pensions demanded by lenders have spared a corrupt, tax-evading elite and heaped an unfair burden on the poorest sections of society.

The streets of Athens are scarred by repeated bouts of violent protest. Banks, fast-food chains and government ministries have been targeted in arson or bomb attacks.

Responding to huge public pressure after a June 17 election, conservative Prime Minister Antonis Samaras has called for tax cuts, extra help for the poor and jobless, a freeze on public sector lay-offs and two more years to bring Greece's deficit under control.

He will miss the two-day EU summit after undergoing eye surgery on Saturday. Government spokesman Simos Kedikoglou said the prime minister would send a letter to EU leaders calling for "a different approach".

"It will assure them that we will honour our commitments and that we want to meet the fiscal consolidation targets, a balanced budget and other targets included in the accord with our European partners," Kedikoglou told 9.84 radio.

But, he added, "Our experience over the past two years shows that the recipe has brought a rapid rise in unemployment and a deepening recession. Accordingly, they have to let us try a different approach. We are seeking room for growth."

Fast losing patience with the slow pace of reform and a litany of broken promises, the country's euro zone partners, led by paymaster Germany, are reluctant to give more leeway.

The Greek wish-list goes far beyond the "adjustments" offered by euro zone partners to make up for weeks of paralysis during two elections since May and a deeper than expected recession.

All sides have played down prospects for any meaningful progress on Greece at the summit, and Athens says it will take its case to European capitals and Washington once Samaras has recovered from surgery to repair a damaged retina.

Greece's delegation to the summit will be led by 83-year-old President Karolos Papoulias. An aide, who declined to be named, said the president would fly economy class.

Successful transplant of patient-derived stem cells into mice with muscular dystrophy

ScienceDaily (June 27, 2012) ? Stem cells from patients with a rare form of muscular dystrophy have been successfully transplanted into mice affected by the same form of dystrophy, according to a new study published June 27 in Science Translational Medicine.

For the first time, scientists have turned muscular dystrophy patients' fibroblast cells (common cells found in connective tissue) into stem cells and then differentiated them into muscle precursor cells. The muscle cells were then genetically modified and transplanted into mice.

The new technique, which was initially developed at the San Raffaele Scientific Institute of Milan and completed at UCL, could be used in the future for treating patients with limb-girdle muscular dystrophy (a rare form in which the shoulders and hips are primarily affected) and, possibly, other forms of muscular dystrophies.

Muscular dystrophies are genetic disorders primarily affecting skeletal muscle that result in greatly impaired mobility and, in severe cases, respiratory and cardiac dysfunction. There is no effective treatment, although several new approaches are entering clinical testing including cell therapy.

In this study, scientists focused on genetically modifying a type of cell called a mesoangioblast, which is derived from blood vessels and has been shown in previous studies to have potential in treating muscular dystrophy. However, the authors found that they could not get a sufficient number of mesoangioblasts from patients with limb-girdle muscular dystrophy because the muscles of the patients were depleted of these cells.

Instead, scientists in this study "reprogrammed" adult cells from patients with limb-girdle muscular dystrophy into stem cells and were able to induce them to differentiate into mesoangioblast-like cells. After these 'progenitor' cells were genetically corrected using a viral vector, they were injected into mice with muscular dystrophy, where they homed-in on damaged muscle fibres.

The researchers also showed that when the same muscle progenitor cells were derived from mice the transplanted cells strengthened damaged muscle and enabled the dystrophic mice to run for longer on a treadmill than dystrophic mice that did not receive the cells.

Dr Francesco Saverio Tedesco, UCL Cell & Developmental Biology, who led the study, said: "This is a major proof of concept study. We have shown that we can bypass the limited amount of patients' muscle stem cells using induced pluripotent stem cells and then produce unlimited numbers of genetically corrected progenitor cells.

"This technique may be useful in the future for treating limb-girdle muscular dystrophy and perhaps other forms of muscular dystrophy."

Professor Giulio Cossu, another UCL author, said: "This procedure is very promising, but it will need to be strenuously validated before it can be translated into a clinical setting, also considering that clinical safety for these "reprogrammed" stem cells has not yet been demonstrated for any disease."

Obama prepping thousands of lawyers for election

OLYMPIA, Wash. (AP) ? President Barack Obama's campaign has recruited a legion of lawyers to be on standby for this year's election as legal disputes surrounding the voting process escalate.

Thousands of attorneys and support staffers have agreed to aid in the effort, providing a mass of legal support that appears to be unrivaled by Republicans or precedent. Obama's campaign says it is particularly concerned about the implementation of new voter ID laws across the country, the possibility of anti-fraud activists challenging legitimate voters and the handling of voter registrations in the most competitive states.

Republicans are building their own legal teams for the election. They say they're focused on preventing fraud ? making sure people don't vote unless they're eligible ? rather than turning away qualified voters.

Since the disputed 2000 presidential election, both parties have increasingly concentrated on building legal teams ? including high-priced lawyers who are well-known in political circles ? for the Election Day run-up. The Bush-Gore election demonstrated to both sides the importance of every vote and the fact that the rules for voting and counting might actually determine the outcome. The Florida count in 2000 was decided by just 537 votes and ultimately landed in the Supreme Court.

This year in that state alone, Obama and his Democratic allies are poised to have thousands of lawyers ready for the election and hope to have more than the 5,800 attorneys available four years ago. That figure was nearly twice the 3,200 lawyers the Democrats had at their disposal in 2004.

Romney has been organizing his own legal help for the election. Campaign attorney Ben Ginsberg did not provide numbers but said the campaign has been gratified by the "overwhelming number of attorneys who have volunteered to assist."

"We will have enough lawyers to handle all situations that arise," he said.

The GOP doesn't necessarily need to have a numerical counterweight to Obama's attorneys; the 2000 election showed that experienced, connected lawyers on either side can be effective in court.

Former White House counsel Robert Bauer, who is organizing the Obama campaign's legal deployment, said there is great concern this year because he believes GOP leaders around the county have pursued new laws to impede the right to vote.

"The Republican Party and their allies have mapped out their vote suppression campaign as a response to our success in 2008 with grass-roots organization and successful turnout," Bauer said. "This is their response to defeat: changing the rules of participation so that fewer participate."

Several states with Republican leaders have recently pursued changes that could make voting more difficult, including key states such as Florida and Ohio, despite objections from voting rights groups that believe that the laws could suppress votes from low-income and minority blocs.

Republicans dispute that the laws are political, pointing to cases of election fraud and arguing that measures like those requiring voters to show identification are simply common sense.

Independent from the Romney team, a conservative group is prepping an Election Day team of its own to combat possible fraud.

Catherine Engelbrecht, president and founder of True the Vote, said the organization hopes to train and mobilize up to one million volunteers this year, many of them to serve poll watchers. One of the group's main initiatives is to "aggressively pursue fraud reports."

"Being a poll watcher is an age-old tradition and we're fortunate that so many volunteers are ready and willing to take a day off, learn what they need to know and help out at the polls," Engelbrecht said. True the Vote already has thousands signed up to help and had 500 trained election workers monitoring the Wisconsin recall vote earlier this month.

"They serve as volunteer guardians of the republic, to ensure that procedures at the polls are in keeping with state law," she said.

It's one of the efforts that have Obama's team fretting. The Democrats fear that anti-fraud activity could get out of hand, with vigilante poll watchers targeting and intimidating voters who may not know their rights.

"We will have the strategy and the resources to address the threat and protect the voter," Bauer said.

The Obama-aligned attorneys, most of whom are not election experts by trade, undergo training and have materials to show them how to help at the polls on Election Day.

Charles Lichtman, who is helping advise the effort in Florida this year after leading it in the last two cycles, first created the Florida Democratic Lawyers Council after the 2000 election, vowing that there would never be a repeat of that disputed vote. He contends Democrat Al Gore would have won the presidency over Republican George W. Bush if a similar legal infrastructure had been in place then.

Lichtman's efforts have since been replicated for other states. He said that is vital to provide voter protection.

"My experience has been that, in every election, the other side has taken drastic measures to try to suppress the vote," Lichtman said. The volunteer organization has not been involved in the 2012 legal disputes so far, though they are monitoring the developments.

Four years ago, the teams of lawyers organized by Obama and Republican candidate John McCain in 2008 went largely unused since the election wasn't very close.

But this year may be different given all the changes to voting laws ? and the closeness of the race in recent polling.

The states with the strictest ID laws require voters to show photo identification before casting ballots. If they don't have proper identification or fail to bring it, they can cast a provisional ballot but must later go to meet with state elections administrators to sort things out before the ballot is counted.

Voting groups see a variety of potential problems, such as how voters are informed of the rule changes, how poll workers handle voters who fail to bring IDs and whether voters are provided adequate notice of the steps they need to take after casting an absentee ballot.

About 30 states have some form of an ID law, with varying methods of implementation.

Legal challenges typically start coming in the weeks before the election, but "litigation has started coming sooner and more vociferously" this year, says Edward Foley, an elections law expert with Ohio State University. That includes lawsuits surrounding Florida's plan to purge ineligible voters from the rolls.

Foley said. "We're in an era of increased litigiousness over the voting process."

He said lawsuits after Election Day may occur only if votes in a battleground state are within the "margin of litigation." That would probably be a difference of just hundreds of votes, a result that would be rare.

Tim Wakefield was 33-36 during his career in June, according to a monthly breakdown by Baseball-Reference.com. The website lumps September and October together, and in that period Wakefield was 33-33.

Phil Niekro ("Knucksie") was 58-43 in June and 50-47 in the September/October period. Hoyt Wilhelm was 21-29 in June and 36-14 in September/October.

September can be plenty warm, of course, so that might skew the September/October figures away from the effect of cold days in October.

In any case, the World Series this year doesn't start until Oct. 24, by which time Dickey and the Mets ? if they get that far ? might have those hand warmers ready just in case.

Dickey is 11-1 with a 2.00 earned-run average and has won his last nine decisions. He's scheduled to start again Sunday against the New York Yankees.

Dickson shines

O'Koyea Dickson, a first baseman with the Dodgers' Class-A Great Lakes (Michigan) team, went two for three with a two-run home run to help the East win, 18-2, in the Midwest LeagueAll-Star game.

Lichen can survive in space: Space station research sheds light on origin of life; potential for better sunscreens

ScienceDaily (June 23, 2012) ? You can freeze it, thaw it, vacuum dry it and expose it to radiation, but still life survives. ESA's research on the International Space Station is giving credibility to theories that life came from outer space -- as well as helping to create better sunscreens.

In 2008 scientists sent the suitcase-sized Expose-E experiment package to the Space Station filled with organic compounds and living organisms to test their reaction to outer space.

When astronauts venture on a spacewalk, hours are spent preparing protective suits to survive the hostile conditions. No effort was made to protect the bacteria, seeds, lichen and algae attached to the outside of the Space Station, however.

"We are exploring the limits of life," explains ESA's Ren? Demets.

Our atmosphere does a wonderful job of protecting life on Earth by absorbing harmful UV rays and keeping temperatures relatively stable.

In contrast, the space samples endured the full power of the Sun's rays. The samples were insulated somewhat by the Space Station but still had to cope with temperatures changing from -12?C to +40?C over 200 times as they orbited Earth.

The samples returned to Earth in 2009 and the results have now been published in a special issue of the journal Astrobiology.

Lichen have proven to be tough cookies -- back on Earth, some species continue to grow normally.

Ren? explains, "These organisms go into a dormant state waiting for better conditions to arrive."

The lichen have attracted interest from cosmetic companies. They can survive the full power of the Sun for 18 months, so knowing more could lead to new ingredients for sunscreen.

Living organisms surviving in open space supports the idea of 'panspermia' -- life spreading from one planet to another, or even between solar systems.

It seems possible that organisms could colonize planets by hitching rides on asteroids. ESA is probing this intriguing theory further on future Station missions with different samples.
